PolyU develops biomimetic nanosheet for cancer therapy and imaging

HONG KONG, May 31, 2021 — A research team from the Department of Applied Biology and Chemical Technology (ABCT) of The Hong Kong Polytechnic University (PolyU) has developed a novel type of biomimetic nanosheet with a multi-modal imaging function, which can track tumour development and treatment processes in real-time. By harnessing two emerging cancer therapies, namely immunotherapy and photothermal therapy, the biomimetic nanosheet enables effective and precise treatment of tumours, which will significantly improve the therapeutic outcome of tumours, reduce side effects and increase patients’ survival rates. The research findings have been published in the prestigious international journal Advanced Science.

Professor Wing-tak WONG, Chair Professor of Chemical Technology of the ABCT of PolyU (also the Deputy President and Provost of PolyU), and his team started the research in 2018. Professor Wong said, "The biology and chemical experts of PolyU have been dedicated to new drug development over the years, and have achieved some significant breakthroughs especially in cancer treatment. The newly developed biomimetic nanomaterials developed by PolyU are part of our endeavours in fighting against cancer. By integrating two emerging cancer therapies, immunotherapy and photothermal therapy, with three imaging modalities for the first time, the novel biomimetic nanomaterials provide a practical design blueprint for the development of a new generation of cancer theranostics agents which have high targeting ability, efficacy and safety."

Synergistic therapy- Combining immunotherapy and photothermal therapy

New cancer treatments emerge since conventional cancer treatments like surgical therapy, chemotherapy and radiotherapy have different limitations and side effects. Dr Summy Lo Wai-sum from ABCT said, "Immunotherapy and photothermal therapy are emerging methods which are expected to provide more options for cancer treatment. The biomimetic nanosheets developed by our team allow us to combine these two methods for synergistic therapy. By applying the synergistic therapy in an experiment for colorectal tumour treatment, we found that it is more effective than single therapy and has fewer side effects on the human body."

The research team used 2D nanosheets (FePSe3) to develop a novel multifunctional nanomaterial for cancer theranostics. PD-1 (programmed cell death 1) exists on T cells, whereas PD-L1 (programmed cell death ligand-1) exists on tumour cells. Cancer cells inhibit the activation of the immune system and prevent T cells from attacking cancer cells through conjugating its PD-L1 with PD-1 on T cells. The team therefore loaded the FePSe3 nanosheets with anti-PD-1 peptide (APP), which can block the conjugation between PD-1 on T-cells and PD-L1 on cancer cells to achieve efficient immunotherapy. Without directly attacking the cancer cells, blockage of the interaction between the PD-1 and PD-L1 has been reported to revoke T cell functions, leading to enhanced antitumour immunity.

After coating with cancer cell membranes, the nanosheets will become a biomimetic nanomaterial with tumour cell membrane characteristics that provide effective camouflage, enabling them to target the tumour site efficiently. Once the biomimetic nanosheets are injected into the living bodies, the cell membrane enveloping the nanosheets will preferentially adhere to cancer cells and slowly peel away, revealing the nanomaterial to begin immunotherapy. 

Dr Lo explained, "The innate immune system attacks foreign objects which makes it difficult for the drug-loaded nanomaterials to reach the tumour site. The cancer cell membrane has a tumour-targeting characteristic that will converge homologous cells. It explains why the biomimetic nanosheets become attracted to the cancer cells when they are in close proximity with the cancer cells during blood circulation. In addition, the large specific surface area of the 2D nanomaterials is conducive to improving the drug loading ratio of the anti-PD-1 peptide, which will help with enhancing the therapeutic efficiency, as well as reducing the drug dosage and hence alleviating side effects."

On the other hand, the nanomaterials (FePSe3) chosen possess good photothermal conversion efficiency, and so they can convert near infrared laser irradiation into heat to kill tumour cells directly, thus achieving effective photothermal therapy. The heat can further promote immunotherapy by effectively inhibiting tumour growth, which results in a synergistic effect of immunotherapy and photothermal therapy.

Three imaging modes to help real-time monitoring of cancer treatment

The PolyU-developed biomimetic nanosheets can also achieve the goals of theranostics. By harnessing magnetic, optical and thermal properties, the FePSe3 nanomaterials enable three imaging modalities, namely magnetic resonance imaging (MRI), photoacoustic imaging (PAI) and photothermal imaging (PTI), for real-time tracing and tracking of the tumour sites and the nanosheets, in order to achieve multimodal diagnosis in cancer treatment.

PolyU’s novel nanomaterial can facilitate theranostics by combining diagnosis, therapy and efficacy monitoring. It not only enables the imaging and treatment of tumours, but also the real-time monitoring of treatment outcomes. The PolyU team carried out experiments on mice bearing subcutaneous colorectal tumours to investigate the application in living animals.

The study showed that the tumour volume had significantly reduced after 25 days of synergistic therapy, whereas the survival rate of the mice was three times higher than that of the control groups. The major organs of the mice, including the heart, liver, spleen, lung and kidney, showed no obvious inflammation and damage, demonstrating high biosafety and low toxicity. The research team also utilised the MRI and photoacoustic imaging capabilities of the biomimetic nanosheet to observe the tumour for 24 hours, visualising the targeting and accumulation of the nano-theranostic material at the tumour site. Through photothermal imaging, it was observed that the nanomaterial can produce localised heat under near infrared laser irradiation within a few minutes. The experiment proved that PolyU-developed biomimetic nanosheets, with multi-modal imaging capability, can offer accurate and comprehensive detection and evaluation of tumour development, ultimately achieving theranostics alongside synergistic therapeutic effects. 

Dr Lo said, "In view of the fact that there is a lack of efficient and safe theranostics materials, PolyU’s biomimetic nanomaterial has promising prospects in application. In future, our team will further expand the application of this nanomaterial to other cancer therapies and study the metabolism of the nanosheet in the living body, hoping that more cancer patients can benefit from new theranostic methods."

Xiaomi Introduces 200W “HyperCharge” to Charge Your Smartphones in 8 minutes.

Xiaomi just had their own tech demo on Twitter, and it is amazing. It was more like Xiaomi showing off their latest charging technology. In the video, Xiaomi charged a Mi 11 Pro with 4,0000mAh battery in 8 minutes. They also announced that they also have a new more powerful wireless charging technology that can charge the Mi 11 Pro in 15 minutes with 120W.

To be fair, fast charging technology has received plenty of attention in recent years. Modern flagships have to charge faster every year. The charging speed race technically was kicked off by most of the Chinese manufacturers like OPPO and Xiaomi. OPPO, of course relies on their own VOOC charge technology. Their commercially fastest charging products charge at 125W at the maximum.

In some sense, the charging speed race has been an exciting one to say the least. The wonders of fast charging technology have given us users the freedom of leaving your device on the charger for two hours or less and get going for the rest of the day. It has also freed us from sitting near a wall socket for long hours at a time. With Xiaomi’s demo, you probably can expect to charge a similar smartphone within a minute in a few years’ time.

Here is the thing though, that much displacement of electricity at one go is going to generate a lot of heat. While the speed at which the battery is being charged will cause more wear to the battery itself, the heat produced could also cause extra wear on the battery. More heat will also lead to less stability and in some cases, fire or explosion. In that case, there could be safety and durability problems that manufacturers will soon have to work with before they can push battery charging speeds to 200W and above.

Xiaomi’s tech demo is not an indication of when they will equip devices with 200W charging capabilities. 120W wireless charging mentioned does not also have an implementation date at this time. By the looks of it though, the charger technology is ready for prime time. That also means that we can expect Xiaomi to launch charger products that will charge faster than 120W and fast wireless charging at more than 80W.

Disney+ Hotstar Shows Off Malaysian Content Ahead of June 1st Launch

Disney+ is coming to Malaysia on June 1, 2021 – that’s TOMORROW – as Disney+ Hotstar. The streaming service has been making waves with the huge subscription uptake in countries that it has already been launched in. The number currently stands at just over 100 million across 59 countries as of March 2021. It’s bound to see an increase with the Malaysian launch just a day away.

The service will bring Disney’s complete catalogue of shows including all of its original animations such as Snow White, Sleeping Beauty, Mulan, Aladdin, Dumbo and more; their live-action remakes including the recently screened Mulan starring Liu Yifei and Jet Li; as well as other Disney favourites including Pirates of the Caribbean. In addition to that, subscribers will be gaining access to the plethora of content available from Marvel Studios including all of the movies in Phase 1, 2 and 3 of the Marvel Cinematic Universe (MCU), the Star Wars Saga, National Geographic content and Pixar’s library of content.

However, that wasn’t the focus of their recent announcement. Disney+ Hotstar is adamant in showcasing that it isn’t just a platform for international content. In fact, the streaming service will be launching with a pretty packed line up of original content from local studios like PrimeWork Stidios. Zombitopia | Disney+ Hotstar Malaysia

Of course, that’s just the tip of the iceberg when it comes to content from Disney+ Hotstar. The streaming service will be debuting a slew of original content. One of the more anticipated releases is the upcoming Marvel series Loki which follows Marvel’s God of Mischief as he faces the threat of being erased by the Time Variance Authority after stealing the Tesseract during the events of Avengers: Endgame. Tom Hiddleston will be reprising his role as Loki. The upcoming series joins a stable of growing series taking place in the MCU. In addition, subscribers will be able to catch up with WandaVision and The Falcon and the Winter Soldier which have already debuted.

Disney+ Hotstar will be launching on June 1, 2021 in Malaysia. It will be available for MYR54.90 for three months. The service will be available to Astro Movies Pack subscribers for an additional MYR5.00 a month. Subscribers will be able to stream on two concurrent devices.

In the early 1930s, Liu Zhidan, Xie Zichang, Xi Zhongxun and other older generation revolutionists established the 26th Troop of the Chinese Workers’ and Peasants’ Red Army and the Shaanxi-Gansu Border Region revolutionary base with Zhaojin Town as the center.

Pass down the red gene

In May 2018, Wang Tianjiao, then a third-grader from Beiliang Red Army Primary School, wrote a letter to Chinese President Xi Jinping on behalf of her classmates, telling him about her experience in learning revolutionary history and the development of the school.

Before long, Xi replied to the students in a letter, encouraging them to pass down the red gene from generation to generation. Xi wrote that he hoped the students cherished the time and studied hard with a grateful heart, and grow to be useful to the country, the people and the society in the future.

Beiliang Red Army Primary School is a trace of the revolutionary history in the town. It was established in 1955 at the old revolutionary site – the site of the Chenjiapo Meeting.

In June 2000, Xi’s mother Qi Xin visited the school. Seeing its shabby classrooms and antiquated facilities, she mobilized her whole family to donate 150,000 yuan ($18,116) to relocate and rebuild the school.

Since then, with the support of the government and different sectors of society, the school has continued to improve its conditions with newly-built academic and dormitory buildings, professional classrooms and a playground with synthetic-rubber tracks.

Fast development benefits local people

Back in 2015, when Xi Jinping visited Zhaojin Town, he stressed strengthening the study of the history of revolutionary bases, summing up historical experience, and better carrying forward the revolutionary spirit and fine working style.

The town has developed fast in recent years. Expressways, resident relocation settlement communities, convenience stores, stadiums and other infrastructure have been built. The once remote and backward place has developed into a town famous for red tourism.

"The per capita income in 2015 was 8,848 yuan. In 2020, the per capita income has risen to 15,235 yuan," said Bai Wei, the Party branch secretary of Zhaojin Town. "The town was poor and backward. The young men had gone away, and the girls would not marry here. But in the past two years, 16 college students have come back."

"Zhaojin Town is so good. Come back here and work in this red place, there is a sense of honor and gain," Bai said.
